A two-step sandwich enzyme immunoassay for rabbit proMMP-3 using monoclonal antibodies

Abstract

A two-step sandwich enzyme immunoassay (EIA) system for the precursor of rabbit MMP-3 (r-proMMP-3) using monoclonal antibodies has been established to quantify the proMMP-3 levels in t he rabbit body fluids. The EIA system takes 150 min for the reactions (120 min for the first and second immunoreactions and 30 min for color reaction at 20°C). Sensitivity for rabbit proMMP-3 was 4.7 ng/ml (140 pg/assay) and linearity was obtained from 17 1,067 ng/ml (0.5 32 ng/assay). The EIA system recognizes only proMMP-3, but not its reactive forms. The levels of proMMP-3 (111 ± 9.3 ng/ml) in the synovial fluids from rabbit osteoarthritic models were higher than those from the normal control animals.
