Gene transfer into macrophage using chitosan nanoparticle modified with mannose

Mannose-modified chitosan was synthesized for receptor-mediated gene delivery to macrophage. In this study we evaluated the efficiency of the cellular uptake and transfection of chitosan/pDNA complexes. Mannosylation to chitosan enhanced the cellular uptake and transfection efficiency. Inhibition assay with mannan indicated mannose-modified chitosan/DNA complexes interacted with mannose receptor on macrophages. Moreover, the effect for activating of macrophages by chitosan/pDNA complexes was also evaluated. The production of NO was promoted by the chitosan/pDNA complexes.
