3D fiber-shaped culture system promotes differentiation of multi potent DFAT cells into smooth muscle-like cells

Here we show that 3D fiber-shaped culture system enhances the differentiation of multipotent DFAT (dedifferentiated fat) cells into smooth muscle-like cells. DFAT cells cultured as 3D cell fiber constructs were induced to differentiate into smooth muscle cells. We found expression of smooth muscle lineage specific markers ASMA ( α-smooth muscle actin) and calponin in the differentiated DFAT cell fibers. The expression of calponin as marker of more mature smooth muscle cells has not been achieved in 2D cultures before. These findings suggest that 3D fiber microenvironment is able to promote more efficient differentiation into smooth muscle-like cells compared to 2D culture.
