Detection of a K-ras point mutation with DNA-conjugated microspheres by surface plasmon resonance

A method for increasing the specificity of detecting a DNA point mutation and the sensitivity of evanescent wave based biosensors was presented. A peptide nucleic acid (PNA) ligand was used to increase the specificity to detect G-G mismatch in K-ras gene affiliated with pancreatic cancer. It was observed that affinity constants between complementary and mismatched targets was of the order of 102 and the largest for PNA interacting with 11mer targets. The results show that change in flow rate has a different effect on the stability of microspheres in a sandwitched geometry that are hybridized to the surface.
